To prepare XLD Agar powder, suspend the required amount in distilled water, heat with agitation until boiling to dissolve completely, then sterilize by autoclaving at 121 °C for 15 minutes. Cool to 45–50 °C and pour into sterile Petri dishes. Do not overheat or re-melt after solidification.
